Around 4,000 people will take to the streets on Sunday 14 October.
The 13.1 mile route takes in the city centre and the countryside along the Exe Valley.
It starts at Exeter Arena at 0900. Runners will then head to the city centre, out past Exeter Central, then up the A379 Valley Road to Stoke Woods, where they turn around and come back.
Road closures and diversions will be in place from 0800.
